چکیده

quick identification of vibrio cholerae in epidemics is   important, on the other hand; conventional methods are time-consuming and   costly. the aim of this study was to develop a rapid, inexpensive and high   sensitivity method for quick identification of vibrio cholerae . for   this purpose we designed a pcr detection based on magnetic nanoparticles for   identification of bacterial dna by pcr dynabead. so we used the biotinylated   probe for binding to dna extracted from vibrio cholera e and other bacterial   species ( salmonella, shigella, pseudomonas, e.coli ) using magnetic   bead isolated with magnetic field, the vibrio genome-specific primers (hlya)   for   pathogen detection (pcr) was used. the results showed specific band was just   for vibrio cholera (pcr positive); therefore designed probe was   specific for vibrio cholera e. according to the findings, this study is   characterized the high sensitivity of pcr using biotin-containing probes for   dna of vibrio cholera in contrast to the traditional methods.

Rapid polymerase chain reaction method for detection of Vibrio cholerae in foods.

The polymerase chain reaction was used to selectively amplify sequences within the cholera toxin operon from Vibrio cholerae O1. Oysters, crabmeat, shrimp, and lettuce were seeded with V. cholerae and then homogenized or washed with alkaline peptone water, followed by short-term (6- to 8-h) enrichment. A detection limit of as few as 1 V. cholerae CFU per 10 g of food was obtained with amplifica...

Rapid Detection of Campylobacter jejuni by Polymerase Chain Reaction and Evaluation of its Sensitivity and Specificity

Introduction: Campylobacter jejuni is one of the most common causes of food poising in humans. Rapid and specific detection of these bacteria has an important role in diagnosis and treatment of infection. The aim of this study was to design a specific PCR for the detection of Campylobacter jejuni. Methods: In this experimental study, oxidoreductase gene from the Campylobacter jejuni was sele...

RAPID DETECTION OF MYCOBACTERIUM TUBERCULOSIS IN CLINICAL SPECIMENS BY POLYMERASE CHAIN REACTION

We investigated the use of DNA amplification by polymerase chain reaction (peR) for detection of Mycobacterium tuberculosis in 300 patients who were suspected of having pulmonary tuberculosis and compared the results with culture results which were performed in parallel with PCR. Two-thirds of each sample was processed for smear and culture by standard methods and one-third was prepared fo...

Detection of luciferase gene sequence in nonluminescent Vibrio cholerae by colony hybridization and polymerase chain reaction.

Bioluminescence is a trait observed among approximately 10% of Vibrio cholerae isolates. We have demonstrated that not only do some strains of V. cholerae produce low levels of light, undetectable by the human eye, but the luciferase gene sequence is present in strains of V. cholerae which emit no detectable light, evidenced by hybridization with a luciferase DNA probe. Comparisons of the amino...

Multiplex Real-time Polymerase Chain Reaction Assays for Simultaneous Detection of Vibrio cholerae, Vibrio parahaemolyticus, and Vibrio vulnificus

OBJECTIVES A multiplex real-time polymerase chain reaction (RT-PCR) method was developed for the identification of three Vibrio species: Vibrio cholerae, Vibrio parahaemolyticus, and Vibrio vulnificus. METHODS Specific primers and probes targeting the hlyA, tlh, and vvhA genes were selected and used for multiplex real-time PCR to confirm the identification of V. cholerae, V. parahaemolyticus,...

Polymerase chain reaction method for the rapid detection of virulent Shigella spp.

Bacillary dysentery, or shigellosis, is a disease of humans in which the colonic epithelium is invaded by bacteria and subjected to inflammatory destruction. The aim of this study was to develop a polymerase chain reaction(PCR) test for detection of virulent Shigella spp.. For this purpose, the primers were designed to amplify a 526-bp internal region of the Shigella spp. icsA gene, which encod...
